Abstract | 「要旨」PGE1およびPGI2の微小血管に対する効果の違いをラット腸間膜にて, 顕微鏡直視下にimage splitting法で計測, 観察し比較した. (1)細動脈に対する直接の拡張作用はPGI2>PGE1にも拘わらず, ノルエピネフリンの細動脈収縮作用に対してはPGE1は抑制するが, PGI2は影響しない. (2)血圧を約20%低下させるように全身投与されたPGE1は逆に細動脈を収縮し, PGI2は影響しない. そして, ノルエピネフリンの細動脈収縮作用をPGE1は亢進しPGI2は抑制するなど, 単に強弱だけでなく, 他の血管作動薬との相互作用の面で効果の変わることが認められるなどプロスタグランディンの循環系への効果が一定しないことを細動脈レベルで実証した. |

Abstract | In the present study, the effects of prostaglandin E1 and I2, were directly observed microscopically and their efficacy were comparatively studied. In the experiment, the mesenteric arterioles (20-30μm) of 6 weeks old SD strain male rats were measured and observed by image splitting method with T.V. monitering. Then, following results were obtained. By topical application ; (1) PGE1 (10-1, 100, 101μg) and PGI2 (10-1, 100, 101μg) produced a dose-dependent dilation ; the magnitude of potency was PGI2>PGE1. (2) Arteriolar responses to 1 μg NE were inhibited for 30 mins. by PGE1 and not inhibited by PGI2. By intravenous application ; (1) PGE1 (3.0 μg/kg/min) elicited arteriolar consrictor responses (max 86.9 ± 3.6% to control) and PGI2 (0.3μg/kg/min) ineffective. (2) Arteriolar responses to 1 μg NE were enhanced for 30 mins. by PGE1 and were inhibited for 5 min. by PGI2. When we use PGE1 and PGI2 in clinical trials, we must consider special qualities of PGE1 and PGI2 and must reduce the side effects of PGE1 and PGI2 to minimum. |
