Abstract | 「要旨」24頭の雑種成犬を使いα-hANPの血行動態に及ぼす影響について検討した. α-hANP投与量により対照群, 0.3μg/kg/min投与群, 1μg/kg/min群, 10μg/kg/min群に分け, α-hANPを60分間投与した. 測定項目は, 血圧, 心拍数, 心拍出量, 血液ガス, 腎動脈血流量, 椎骨動脈血流量, 尿量で, 対照値, 投与30分, 投与60分, 投与中止後60分の4時点で測定を行った, 平均血圧は各α-hANP投与群とも有意な低下を示した. 心拍数は各群とも変化は認めなかった. 心拍出量は0.3μgでは減少傾向を示し, 1μg, 10μg群では有意な減少が認められた. PaO2は各α-hANP投与群で有意な低下が認められた. 尿量は各α-hANP投与群で投与中は増加が認められた. 椎骨動脈血流量は各α-hANP投与群で減少したが, 血圧低下, 心拍出量減少にもかかわらず腎動脈血流量は比較的一定に保たれており, α-hANPが腎動脈に作用し血管拡張を起こし血流を維持したためと推測された. |

Abstract | Twenty-four mongrel dogs were anesthetized with pentobarbital and anesthesia was maintained with nitrous oxide-air-enflurane. Arterial carbon dioxide tension was kept about 40 mmHg with a Harvard ventilator. A catheter was inserted to femoral artery to measure blood pressure and heart rate, and a Swan-Ganz catheter was advanced to pulmonary artery from femoral vein to measure cardiac output by thermodilution technique. Probes of ultrasonic transit flowmeter were attached to left vertebral artery and, renal artery to measure the blood flow of eachorgan. Urine volume was measured continuously. Dogs were divided into four groups:1) control group in which normal saline was infused at the rate of 20 ml/hour. 2) small dose group in which α-hANP was infused at the rate of 0.3 μg/kg/min. 3) moderate dose group in which α-ANP was infused at the rate of 1 μg/kg/min. 4) high dose group in which α-hANP was infused at the rate of 10 μg/kg/min. Increasing the infusion rate of α-ANP, both blood pressure and cardiac output were decreased progressively. At the same time urine volume was also increased. progressively. Simultaneous decrease of vertebral arterial blood flow was occurred with the decrease of cardiac output, whereas the renal blood flow remained at the control level. It would be suggested that vasodilatory action of α-hANP was specifically found in renal artery and would contribute to cause significant diuresis. |
