Abstract「要旨」交感神経末端遊離促進性β受容体を介するnoradrenaline(NA)遊離反応が, Adriamycin心筋症心不全において, どのようになっているかを検討するため, Adriamycin心筋症rabbit[日本白色種の雄, 体重2.5±0.04s:投与量1mg/kg, 週2回×8週間静注]を用い, pentobarbital麻酔下(70mg/kg bolus i.v., 15mg/kg/hr持続注入), adrenaline(0.06μg/kg/min)持続静注時の血漿NA濃度を測定(HPLC+THI法), 無処置群(Vehicle群)と比較した. その結果, (1)adrenalineによる血漿NA濃度の増加は, Vehicle群(n=7)に比して, adriamycin処置群(n=7)にて, 有意に小であった. (2)Vehicle群において, adrenalineによる血漿NA濃度の増加はpropranololによってほぼ完全に抑制された. Adriamycin心筋症心不全において, 遊離促進性β受容体を介するNA遊離反応は, 低下していることが示唆された.

AbstractWe investigated the role of release-enhancing β-adrenoceptors in noradrenaline release in rabbits with cardiomyopathy produced by adriamycin (1 mg/kg, i.v., twice a week for 8 weeks). Plasma noradrenaline levels were measured before, 30 min and 60 min after the start of continuous intravenous administration of adrenaline (0.06 ug/kg/min) in adriamycin-treated and vehicle treated rabbits in anesthetized subjects. In anesthetized condition, adrenaline increased plasma noradrenaline levels in vehicle-treated rabbits. However, in the adriamycin-treated rabbits, adrenaline had no effect on the plasma noradrenaline level. Pretreatment with propranolol (0.2 mg/kg,bolus i.v.+0.1 mg/kg/hr, continuous infusion) almost completely abolished the rise in plasma noradrenaline level associated with adrenaline infusion in vehicle-treated rabbits. These results suggest that in rabbits with adriamycin-induced cardiomyopathy, the noradrenaline release from the sympathetic nerve endings via the activation of release enhancing β-adrenoceptors is reduced.
