Abstract | 「要旨」エンドトキシン(ET)の作用によって生じた肝微小循環変化に対してPentoxifylline(POF)がいかにこれを修飾するかラット摘出肝灌流法において研究した. ET投与により, 胆汁分泌量は減少し, 肝の類洞内血管容積は減少し, 血管外アルブミン分布容積は増加した. しかしPOF投与によって胆汁分泌量の減少は抑制され, 肝の血管外アルブミン分布容積は回復した. ET投与1時間前のPOF処置によってET投与後の灌流圧, 肝の類洞内血管容積, 血管外アルブミン分布容積およびシャント率に変化は見られず, 胆汁分泌量のみが, ET投与2時間後に減少する傾向を示した. すなわちPOFは, ETによる肝微小循環変化を防止し, また一部改善する事が示された. その作用はETによるサトカイン遊離を抑制するためではないかと想像された. |

Abstract | The microcirculatory effect of pentoxifylline (POF) was observed before and after administration of endotoxin (E. coli 0111, B4, Sigma) in isolated rat liver. Following endotoxin (2 mg・dl-1) administration, bile flow and sinusoidal volume decreased and Disse's space increased significantly. After POF (6 mg・dl-1) administration, the bile flow remained unchanged but Disse's space recovered toward the initial value. Pretreatment with POF, which was administered one hour before the endotoxin infusion, caused no significant changes in the mean perfused pressures, sinusoidal volume, Disse's space, or shunt ratio, but did tend to decrease the bile flow after two hour's administration of endotoxin. This study showed that POF improved microcirculatory changes in the liver, which had been impaired after endotoxin infusion and also that prevent those changes could be prevented by pretreatment with POF. It was presumed that the release of cytocaine from hepatic cells (probably Kupffer cells) caused by endotoxin might have been inhibited by POF. |
