Abstract | 「要旨」ペントバルビタール麻酔下のイヌ10頭に対して超短時間作用性β遮断薬ONO-1101(1101)を投与し, その心血行動態に及ぼす影響ならびに投与中止後の回復過程を観察した. 10(S1), 30(S2), 100(S3)μg/kg/minの速度で1101を順次段階的に投与し, S3での計測後投与を中止, 10(S4), 20(S5), 30(S6), 分後にも観察を行った. 1101の投与により用量依存的な強い陰性変時作用が観察された. 血圧には変化はなく, rate-pressure productは減少した. 心拍出量は減少したが, 左室1回拍出係数は増加し, 体血管抵抗は上昇した. 左冠動脈回旋枝血流量は減少し, 冠血管抵抗は上昇した. 投与中止後, 心血行動態はただちに回復しはじめ, 20〜30分後には大部分のパラメータは対照値に近く回復した. 1101の安全域は広く, 超短時間作用性で調節性に優れることから今後臨床での有用性が期待される. |

Abstract | The effects of an ultra short-acting β-blocker (ONO-1101) on systemic and coronary hemodynamics were studied in 10 pentobarbital-anesthetized mongrel dogs. After control measurement, the drug was infused intravenously for 30 min at the rate of 10, 30 and 100 μg/kg/min, respectively. Hemodynamic measurements were performed at the end of each dose administration, and 10, 20 and 30 min following termination of the last dose. The drug produced dose-dependent decreases in heart rate, rate-pressure product, cardiac output, left circumflex coronary blood flow and LV dP/dt max except left ventricular stroke volume index, which increased dose-dependently. Both systemic and coronary vascular resistances were increased. After termination of the drug infusion, β-blockade of the drug rapidly returned to the baseline values within 20-30 min. It is concluded that the newly developed ultra short-acting β-blocker may be useful in clinical settings because of its short duration of action and easy control. |
