Abstract | 「要旨」ウサギにおいてischemic preconditioning(PC)のメカニズムとして, adenosine A1 receptor, protein kinase Cの他に, PC中に交感神経末端より放出されるnoradrenaline(NA)によるα1B receptor刺激が重要である. これらのブロッカーは単独で, PC効果をブロックできることから, 情報伝達系は, 心筋細胞レベルで相互に関連しているとされる. 我々は, 交感神経レベルでもこれらのメカニズムが起こっているという仮説を考えた. 日本白色種ウサギ(オス, 約2kg)の心臓前側壁枝の5分虚血・5分再灌流のPC前後に, microdialysis法を用い, 心筋間質内NA濃度をHPLCと電気化学法にて測定した. PC中に生ずる心筋間質内NA濃度の上昇が, adenosine A1 receptorおよびprotein kinase Cのブロッカー(8-p-sulfophenyl theophylline : 8-SPT, 7.5mg/kg, i.v., polymyxin B : PB, 24mg/kg, i.v.)でブロックされるか否かを検討した. |

Abstract | Many mechanisms such as α1B-adrenoceptor, adenosine A1 receptor and protein kinase C involve in the ischemic preconditioning (PC) effect. It is considered they are cross-talking each other at the level of myocytes. However, the cross-talk may occur at the level of sympathetic nerve endings. We hypothesized that the increase in cardiac interstitial noradrenaline (NA) by PC is modified by the activation of adenosin receptors and protein kinase C. Methods : In 19 open chest Japanese white male rabbits anesthetized with pentobarbital, cardiac interstitial NA levels in the area at risk were measured before and during PC of 5 min ischemia and 5 min reperfusion in the presence or absence of adenosine blocker (8-p-sulphophenyl theophylline, 7.5mg/kg), protein kinase C inhibitor (polymyxin B, 24mg/kg) using microdialysis technique and HPLC. Results : PC increased cardiac interstitial NA levels and this increase was inhibited by adenosin blocker or protein kinase C inhibitor. Conclusions : PC increases cardiac interstitial NA through activation of adenosine receptor and protein kinase C. |
