| Abstract | Isolated rat heart modelを用いて, 潅流液であるKrebs−Henseleit bicarbonate bufferの中に, Human SODを各種濃度にて添加し, Langendorff mode下, 左前下行枝を10分間閉塞, その後3分間のReperfusionを行い, Reperfusion−induced arrhythmiaを作成し, 各種不整脈の発生頻度, 持続時間について検討した. その結果, 心室細動および非可逆性心室細動の発生頻度において10万unit／lから効果が認められ, 30万unit／lを至適濃度とするbell−shaped dose response curveを示すことが判明した. しかしながら, その作用機序に関しては, 今後の検討を要するものと思われた. |

| Abstract | Using the isolated rat heart preparation, the effects of Human SOD upon reperfusion−induced arrhythmia were evaluated in this study. Under the condition of Langendorff perfusion with Krebs−Henseleit bicarbonate buffer containing various concentration of Human SOD, left anterior decending coronary artery was occluded for 10 min to produce regional ischemia. After this, reperfusion was performed by releasing the occluder. Epicardial ECG was analysed in terms of incidence of ventricular fibrillation （VF） and the duration of VF. Human SOD had the bell−shaped dose response curve and its optimal dose was 30×104 unit／l to prevent the reperfusion−induced arrhythmia. It was concluded that Human SOD has the beneficial effect upon reperfusion−induced arrhythmia when 30×104 unit／l of optimal dose was employed. |
