Abstract | 「要旨」イソフルランの血管平滑筋収縮作用機序を解明するため, カルシウム感受性蛍光色素Fura PE3を用いた細胞内カルシウム濃度と張力の同時測定, および45Ca2+を用いた細胞外カルシウムの流入量の測定を行った. 内皮を除去しFura PE3を負荷したラット大動脈輪状標本をリアノジン(20μM)で処置, ベラパミル(10μM)存在下, フェニレフリン(0.3μM)で収縮させ, イソフルラン(1.2-3.5%)を投与, 細胞内カルシウム濃度と張力の変化を検討した. その結果, 張力, 細胞内遊離カルシウム濃度とも, イソフルラン2.3%および3.5%で有意に増加した. また, 内皮除去ラット大動脈条片を45Ca(1μM), ニフェジピン(10μM)を含むHEPES緩衝液に浸漬し, フェニレフリン(0.3μM), イソフルラン(1.2-3.5%)を投与して細胞外からのカルシウム取り込みを測定すると, カルシウム取り込み量はイソフルラン1.2%および2.3%でイソフルラン非投与群に比し有意に増加した.これらの結果から, イソフルランが受容体作動性カルシウムチャネル(ROC)を介した細胞外カルシウム流入を増加させることが示唆された. |

Abstract | This study attempted to clarify the mechanism of isoflurane-induced contraction of vascular smooth muscle (VSM) by measuring the intracellular free calcium concentration ([Ca2+]i) in and 45Ca2+ uptake by rat aortic VSM in the presence of a supramaximal dose of one of the L-type calcium channel blockers, verapamil and nifedipine. Endothelium-denuded rat aortic rings were loaded with the calcium-sensitive fluorescent dye, fura PE3, in order to measure F340/F380 as an indicator of [Ca2+]i. Isoflurane (2.3% and 3.5%) increased the tension and [Ca2+]i of phenylephrine (0.3μM) -contracted aortae in the presence of verapamil (10μM) even after treatment with ryanodine (20μM). Isoflurane (1.2% and 2.3%) enhanced the phenylephrine-induced uptake of 45Ca2+ by endothelium-denuded aortic strips in the presence of nifedipine (10μM). These results indicate that isoflurane enhances phenylephrine-induced Ca2+ influx through channels resistant to L-type channel blockers. |
